Continuous optical in‐line glucose monitoring and control in CHO cultures contributes to enhanced metabolic efficiency while maintaining darbepoetin alfa product quality

Great efforts are directed towards improving productivity, consistency and quality of biopharmaceutical processes products. One particular area is the development new sensors for continuous monitoring critical bioprocess parameters by using online or in-line systems. Recently, we developed a glucose biosensor applicable in single-use, long-term mammalian cell bioreactors. Now, integrated this sensor an automated feeding system capable maintaining stable levels, even at very low concentrations. We compared fed-batch feedback both (< 1 mM) high (40 levels with traditional batch culture methods, focusing on glycosylation glycation recombinant protein darbepoetin alfa (DPO) produced CHO line. evaluated growth, metabolite product concentration under different strategies show that feeding, has no harmful effects DPO quantity quality. conclude our tight level control throughout extended bioprocesses potential to improve performance where constant maintenance critical.
